Would I be able to request an exact room when cooking, for example room number 1351E as a friend has previously stayed there and said it was great.

It is fine to request a specific room when booking, although this cannot always be guarenteed. Some hotels are fine about booking a specific room, others are not. All this is usally subject to hotel size and also hwo far in advance the booking is.
